Guide RNAs and SNPs for nanopore enrichment

In which part of the gRNA sequence is it most important to not get SNPs? I know the PAM site cannot have any SNPs but what about other ones? And secondly, are there any positions in the gRNA sequence where it is acceptable to have SNPs?
